NMR allows one to study ligand−protein interactions, either by exploiting effects of binding on the linewidths (1/T 2 or 1/T 1ρ ) of prospective drugs, or by
observing effects on the lifetimes of long-lived states (LLSs). The latter
method requires very low protein−ligand ratios, thus saving on costly
proteins. Screening inhibitors of urokinase-type plasminogen activator (uPA)
which is a prototypical target of cancer research, with a concentration of only 10 μM, allowed us to determine a dissociation constant K D = 180 ± 20 nM for a strong inhibitor. The more cumbersome surface plasmon resonance
method gave K D = 157 ± 39 nM.
